Yorkshire set the away side 186 runs to win with Joe Root top-scoring on 55. Overseas bowler James Faulker took 3-27.
Lancashire started their reply well with Liam Livingstone (32) and Ashwell Prince (27) but Buttler hit the winning run off the final ball of the innings to wrap up an enthralling tie.
Lancashire needed 32 off final two overs but wicketkeeper Buttler kept his cool to seal an important and memorable victory.
